摘    要: 目的 研究鸡新城疫病毒对血管的生长抑制作用。方法 采用小鼠肺癌转移模型观测鸡新城疫病毒(NDV)对癌细胞转移的影响及组织内微血管的生长情况;用体外药物敏感实验(MTT)检测NDV在不同剂量和作用时间下对人血管内皮细胞ECV304细胞的增殖抑制效应;用PI和Hoechst33342进行荧光染色检钡4NDV作用后细胞有无凋亡改变及其形态学变化。结果 NDV在体内可以有效抑制小鼠发生癌转移时组织内微血管的生成,在体外可以抑制ECV304细胞的增殖并导致较为轻微的凋亡现象。结论 体内外实验均表明NDV可以抑制肿瘤发生时微血管的生长,这一作用通过抑制血管内皮细胞的增殖而非直接的细胞毒作用实现,该作用机制可能与某种特定病毒蛋白有关,尚待进一步研究。

Abstract:Objective To study the effect of new castle disease viru (NDV) on growth inhibition of vein in vivo and vitro.Methods Studying the effect of NDV on transfer of cancer cells and growth of capillary vessel with lung cancer transfer model of mouse;NDV were incubated with human umbilical vein endothelial cells ECV304 cells in a dose- and time-dependent manner. Anti-proliferation effects were measured by MTT assay, and apoptosis was determined by PI and Hoechst 33342 staining.Results NDV could inhibit the growth of capillary vessel in vivo, could also inhibit the growth of ECV304 cells in a dose- and time-dependent manner. The apoptosis of ECV304 cells could be induced by NDV indistinctively.Conclusion It was indicated by vivo and vitro experiment that NDV could inhibit the growth of capillary vessel, and this function be fulfilled though inhibiting the growth of ECV304 but kill the cells directly. What's more, this function may be induced by some proteins in NDV possibly.
