眼针对脑缺血再灌注模型大鼠ERK1/2信号转导通路影响的实验研究

摘    要: 目的:探索脑缺血再灌注损伤后ERK1/2信号转导通路的激活情况,研究眼针疗法对该通路的影响。方法:应用线栓法复制出脑缺血再灌注损伤大鼠的模型,采用眼针治疗。再灌注24 h后处死取材,采用Western-blot方法对各组大鼠海马组织中ERK1/2及p-ERK1/2的表达进行检测。结果:眼针疗法能够显著上调脑缺血再灌注损伤后大鼠海马组织中ERK1/2、p-ERK1/2蛋白的表达,与模型组比较,有统计学意义。结论:眼针疗法能够显著上调脑缺血再灌注模型大鼠ERK1/2及p-ERK1/2的表达;眼针疗法的脑保护作用可能通过激活ERK1/2信号转导通路而实现。

Abstract:Objective To explore ERK1/2 signaling pathway activation after cerebral ischemia reperfusion injury,to study the effect of eye-acupuncture therapy on the ERK1/2 pathway.Methods string thrombus method was used to replicate the model of cerebral ischemia reperfusion injury rat,eye-acupuncture therapy was applied thereafter.After reperfusing 24 hours,the rats was killed,the ERK1/2 and p-ERK1/2 expression in the rat’s hippocampus were detected by Western-blot method.Results The reduced expression level of ERK1/2,p-ERK1/2 protein in hippocampus of cerebral ischemia-reperfusion injury rat was improved through adopting eye acupuncture therapy,compared with model group.Conclusion Eye-acupuncture therapy could significantly improve the expression level of ERK1/2 and p-ERK1/2 restrained by cerebral ischemia-reperfusion injury in rat hippocampus;eye-acupuncture therapy could make cerebral protected through activating ERK1/2 signaling pathway.
