神经干细胞与骨骼肌细胞的共同培养

摘    要: 目的 观察神经干细胞分化形成的运动神经元能否与骨骼肌细胞形成功能性突触。方法 分离培养小鼠神经干细胞及骨骼肌细胞,将神经干细胞与骨骼肌细胞进行共同培养。用免疫荧光方法检测神经干细胞的分化情况及其与骨骼肌细胞形成突触结构的情况,用药理学方法检测突触结构的功能性。结果 神经干细胞在与骨骼肌细胞的共同培养中分化为运动神经元样细胞,表达胆碱能细胞标志物,可见乙酰胆碱受体沿神经元轴突排列。药理学实验证明功能性突触的存在。结论 神经干细胞在与骨骼肌细胞的共同培养中分化为运动神经元,并能与骨骼肌细胞形成功能性突触结构。

Abstract:Objective To explore the formation of functional neuromuscular synapse in a coculture system of neural stem cells and muscle skeletal cells.Methods Neural stem cells and skeletal muscle cells were primary isolated from mouse.Then they were put into a coculture system.The differentiation of neural stem cells and the formation of neuromuscular synapse were observed via immunofluorescence microscope.Pharmacological approaches were used to test the functional characters of the neuromuscular synapse.Results Motor neuron-like cells were observed in the cocultures.Cholinergic cell were also detected.AchR clusters were observed arranged along neuronal process.Pharmacological approaches showed the function of the synapse.Conclusion Motor neurons differentiated from neural stem cell and skeletal muscle cocultures could form functional synapse with skeletal muscle cells.
