先兆子痫患者蛋白 Z 水平及其内含子 FG79A 基因多态性的实验研究

摘    要:目的探讨先兆子痫患者血浆蛋白z(PZ)水平及其内含子FG79A基因多态性的发生率与先兆子痫之间的相关性和临床意义。方法对先兆子痫组160例和正常妊娠对照组162例检测PZ水平并采用PCR和限制性内切酶片段长度多态性方法并结合基因测序检测PZ内含子FG79A基因多态性,同时检测两组D-D、FDP、AT:A的水平,观察两组各项指标变化情况。结果先兆子痫组PZ水平显著低于正常妊娠对照组(1.57±0.32mg/Lvs2.12±0.35mg/L,P〈0.05);两组人群均存在PZ内含子FG79A基因多态性。先兆子痫组GG型、GA型和AA型分别占19.38%、53.75%和26.87%,G、A等位基因频率分别为46.25%和53.75%;正常妊娠对照组GG型、GA型和AA型分别占17.90%、54.94%和27.16%,G、A等位基因频率分别为45.37%和54.63%,两组基因型和等位基因频率分布无显著差异(均P〉0.05);先兆子痫组D-D、FDP水平明显高于正常对照组(5.15±1.03μg/ml vs0.58±1.32μg/ml,P〈0.05;19.46±3.28μg/ml vs 2.93±1.92μg/ml,P〈0.05),而AT:A水平无显著差异(99.12±13.5%vs100.8±10.6%,P〉0.05)。结论先兆子痫患者存在PZ内含子FG79A基因多态性,但该基因多态性与疾病的发生关联不大,然而该疾病与Pz水平、D-D、FDP水平却有一定的相关性。

Abstract:Objective To investigate the level of PZ and the distribution of PZ intron F G79A polymor- phisms in patients of preeclampsia and clinical significance. Method The test group was consisted of 160 pa- tients of preeclampsia, the control group was consisted of 162 women with normal pregnancy. The PZ intron F G79A polymorphisms were studied by PCR and restriction fragment length polymorphism. Meanwhile, D-D, FDP, AT Ⅲ:A and PZ level were tested. Observing changes in two groups. Results The level of PZ in test group was lower than that in control group( 1.57 ±0. 32 mg/L vs 2. 12 ± 0. 35 mg/L,P 〈 0. 05 ). The PZ in- tron F G79A polymorphisms were found in two groups. The percentages of GG/GA/AA were 19.38%/ 53.75%/26. 87% in test group. The frequencies of G and A alleles were 46.25% and 53.75%. The percent- ages of GG/GA/AA were 17.90%/54.94%/27.16% in control group. The frequencies of G and A alleles were 45.37% and 54.63%. There were no significant difference in PZ intron FG79A genotype and alleles fre- quencies distribution between test group and control( P 〉 0. 05 ). The levels of D-D and FDP in test group were higher than those in control group ( 5.15 ± 1.03 μg/ml vs 0. 58 ± 1.32 μg/ml, P 〈 0. 05 ) ; ( 19.46 ± 3.28 μg/ml vs 2.93 ± 1.92 μg/ml,P 〈0.05) ,there was no difference in ATm :A between two groups(99.12 ± 13.5 % vs 100. 8 ± 10. 6%, P 〉 0. 05 ). Conclusion PZ intron F G79A polymorphisms are existed in pa- tients of preelampsia,but its polymorphisms were not associated with the disease. While the decreased PZ lev- el, increased D-D and FDP levels may be associated with the preeclampsia.
