半乳糖化磁性白蛋白阿霉素纳米粒与阿霉素对大鼠的毒性比较

摘    要:目的 比较研究半乳糖化磁性白蛋白阿霉素纳米粒 (AGMAN)和阿霉素 (ADM )对大鼠的毒性作用。方法 将AGMAN和ADM分为几个不同的剂量组 ,观察静脉给药后的两种药物的急性LD5 0 ,同时将未死亡的动物进行肝肾功能 ,心肌酶学的检查 ,观察其改变。结果 半乳糖化磁性白蛋白阿霉素纳米粒比阿霉素的LD5 0显著的提高。动物的肝肾功能损害明显减轻。结论 AGMAN具有很安全的用药范围 ,对动物的主要器官及全身的毒副作用相对ADM也有明显的减轻

Abstract:Objective To study the toxicity of Adriamycin contained by galactosed magnetic albumin nanoparticle (AGMAN) in rats.Methods 150 animals were divided into 15 groups randomly.8 teams were gived corresponding dose of AGMAN;6 teams were gived corresponding dose of Adriamycin(ADM) and one team rats were gived asepsis water in vein.Then detecting LD50 of the two drugs by the Bliss method;Observing blood biochemical (function of the liver and kidney;enzyme of cardiac muscle),Observing the irritability of the AGMAN;And observing the difference of the tow kinds drug by the SPSS.Results The LD50 of AGMAN was 515.50 mg/kg,and the LD50 of ADM was 10.38 mg/kg.Compared to ADM,AGMAN effectively reduced the changes of the blood biochemistry.The spleen remained the same in both.We observed the anaphylaxis on one of the 20 rats.Conclusion AGMAN significantly diminished the toxicity of adriamycin in the important organs of the body.The LD50 of AGMAN was improved greatly than that of ADM.It is a new type of anticancer drug in the foreground in the future.
