刺五加多糖对异基因骨髓移植小鼠免疫功能重建的影响

摘    要:本文用抗Thy-1.2单克隆抗体和补体在体外特异地去除C_(57)BL/6(H—2~b)小鼠骨髓中的T细胞,成功地对致死量照射的BALB/c(H—2~d)小鼠进行了异基因骨髓移植。移植后55天,异基因骨髓移植小鼠的免疫功能明显低于正常小鼠。在移植后45天,用每日100mg/kg剂量的刺五加多糖经腹腔连续给药10天,小鼠对胸腺不依赖抗原TNP-Ba的PFC反应以及对ConA和LPS增殖反应均较对照小鼠明显增加,但对胸腺依赖抗原SRBC的PFC反应以及BSA诱导的DTH反应无明显改善。

EFFECT OF POLYSACCHARIDE OF ACANTHOPANAX SENTICOSUS (PAS) ON IMMUNOLOGICAL RECONSTITUTION OF ALLOGENEIC BONE MARROW TRANSPLANTATION IN MICE

Abstract:In this communication the anti-Thy-1.2 monoclonal antibody plus complement were used to purge T cells from allogenic bone marrow. The T-cell-depleted allogeaeic bone marrow successfully reconstituted the immunological functions of the lethally irradiated BALB/c mice.On the forty-fifth day after allogeneic bone marrow transplantation the immunologically reconstituted BALB/c mice were injected with PAS at dosage of 100mg/kg/day i.p for 10 days. The PFC response to thymus-independent antigen TNP-Ba and the mitogenic response to ConA and LPS were significantly increased in drug-receiving group but no increase of PFC response to thymus-dependent antigen SRBC and DTK response induced by BSA was observed.
