摘    要:目的:探讨舒心通脉片改善异丙肾上腺素介导的心肌缺血的作用机制与其抑制含Nox4的NADPH氧化酶介导的氧化应激。方法:研究舒心通脉片对异丙肾上腺素诱导的心肌缺血大鼠心电图和左心室Nox4表达水平的影响,以及大鼠给药后血清中乳酸脱氢酶(lactate dehydrogenase, LDH)、磷酸肌酸激酶(creatine phosphokinase, CK)、超氧化物歧化酶(superoxide dismutase, SOD)和丙二醛(malondialdehyde, MDA)浓度的变化。结果:舒心通脉片可抑制异丙肾上腺素介导的心肌缺血大鼠中ST段位移值的增加,降低血清中LDH和CK水平,升高SOD活性,降低左心室中Nox4的表达水平。结论:舒心通脉片可改善异丙肾上腺素诱导的心肌缺血,其机制可能与降低Nox4介导的活性氧的产生和升高SOD活性有关。

Abstract:OBJECTIVE To investigate the improving mechanisms of Shuxintongmai tablets on myocardial ischemia induced by isoprenaline and oxidative stress induced by NADPH oxidase-containing Nox4.METHODS Isoprenaline-induced model of myocardium ischemia was used to test effects of Shuxintongmai tablets on electrocardiogram, concentrations of LDH, CK, SOD and MDA in plasma, and expression of NADPH oxidase-containing Nox4 in left ventricular tissues.RESULTS Shuxintongmai tablets reduced ST displacement, decreased contents of LDH and CK in plasma, and expression of Nox4 in left ventricular tissues, and increased level of MDA in plasma.CONCLUSION Shuxintongmai tablets could alleviate myocardial ischemia in isoprenaline-induced model of myocardium ischemia, the mechanism of which is related to reduction of reactive oxygen species induced by NADPH oxidase-containing Nox4 and elevation of SOD activity.
