卡泊三醇对小鼠黑素瘤B16细胞增殖及凋亡的影响

摘    要:目的探讨不同浓度卡泊三醇(CPT)对小鼠黑素瘤B16细胞增殖及凋亡的影响。方法不同浓度CPT作用于体外培养的小鼠黑素瘤B16细胞24h,在倒置显微镜下观察细胞密度及形态,采用MTT比色法和AnnexinV/PI染色法分别检测细胞增殖抑制率和凋亡率。结果与对照组相比,CPT浓度为1,10μg/mL组中B16细胞密度及形态无明显变化,也无增殖抑制作用(P>0.05)。而当CPT为102,103μg/mL时,细胞密度减少,形态由多角形变为略圆钝,甚至皱缩,对细胞增殖呈剂量依赖性的抑制作用(P<0.05)。CPT为1,10,102μg/mL时对B16细胞无凋亡诱导作用(P>0.05),而103μg/mL时则明显促进细胞凋亡(19.17±0.31)%,差异有统计学意义(P<0.05)。结论 CPT≤10μg/mL对B16细胞密度形态、增殖抑制率和凋亡率无明显影响;CPT≥102μg/mL能改变B16细胞形态,并呈浓度依赖性地抑制其增殖;而当CPT≥103μg/mL时能诱导B16细胞凋亡。

Abstract:Objective To investigate the effects of different concentrations of calcipotriol(CPT) on proliferation and apoptosis of mouse melanoma cells B16.Methods Mouse melanoma cells B16 in cultured vitro were treated with different concentrations of CPT for 24h,then the density and morphology were observed by inverted microscope,the proliferation and apoptosis rate were detected by MTT and Annexin V/PI staining kit,respectively.Results Compared with the control group,there was no significant effects on the morphology,density and proliferation rate by CPT at 1,10 μg/mL in B16(P>0.05).When its concentration was 102 and 103μg/mL,the density of cells decreased and the shape became slightly blunt,even shrinked;the proliferation of B16 was also inhibited dose-dependently(P<0.05).There was no significant effects on the apoptosis rate by CPT at 1,10,102μg/mL in B16(P>0.05).Whereas at 103μg/mL,the apoptosis increased sharply(19.17±0.31)%,with a significant difference(P<0.05).Conclusion There was no significant effects on the morphology,proliferation and apoptosis by CPT≤10μg/mL.CPT≥102μg/mL changed cell shape and produced a dose-dependent inhibition on proliferation.CPT≥103μg/mL could induce apoptosis in B16 cell.
