海绵体上抗肿瘤放线菌的分离筛选及菌株Streptomyces sp.A01059的发酵条件优化

摘    要:目的分离筛选抗肿瘤活性放线菌,并提高菌株Streptomycessp.A01059抗肿瘤活性物质的产量。方法以抗稻瘟霉,MTT法为筛选模型,对分离自海南岛周边海域海绵样品的放线菌进行筛选,并对菌株Streptomycessp.A01059的发酵培养基组成及培养条件进行优化。结果筛选获得8株抗肿瘤活性较好的菌株,优化出菌株A01059发酵的最佳碳源,氮源,盐度,pH值及接种量等。结论优化条件下,菌株A01059发酵液稀释100倍时,对人肝癌细胞SMMC-7721,小鼠肉瘤细胞S180,人正常肝细胞L-20的抑制率分别为80%,81%,12%。

Abstract:Objective To isolate and screen the antitumor actinomycetes and increase the yield of the bioactive metabolite produced by Streptomyces sp.A01059.Methods Based on the models of Pyri-(cularia) oryzae and MTT,antitumor actinomycetes were screened from the marine sponges collected in the coast areas of Hainan Island.The fermentation media and conditions of Streptomyces sp.A01059 were studied,too.Results Eight strains of antitumor actinomycetes were obtained.The carbon source,nitrogen source,salinity,pH value and incubation volume and so on were optimized.Conclusion Under optimized conditions,the inhibitory rates on human liver cancer cell SMMC-7721,rat sarcoma cell S_(180) and human liver cell L-20 were 80 %,81 %,12 %,respectively,when the fermentation broth was diluted 100 times.
