摘    要:目的:建立铁皮石斛带节茎段为外值体的快速繁殖体系,保护和合理利用铁皮石斛资源.方法:以铁皮石斛(Dendrobium officinale Kimura et Migo)的带节茎段作外植体,比较不同培养基、激素等因素对铁皮石斛诱导、增殖及生根的影响.结果:最适茎段诱导腋芽的培养基为1/2MS+ 2.0mg L-1 6-BA+ 0.2mgL-1 NAA,腋芽诱导率可达93.3%;最适丛生芽增殖的培养基为1/2MS+ 1.5mgL-1 6-BA +0.5mgL-1 NAA,平均增殖系数达到5.46;最适的生根培养基是1/2MS+0.5mgL-1 IBA +0.5mgL-1 NAA+ 0.2% AC,组培苗生根多,生根率达到72%.以上各种培养基琼脂粉浓度均为0.8%,蔗糖浓度均为2%,pH5.6 ~5.8.同时研究发现,组培苗的移栽基质以细木屑为优,具有疏松透气、排水良好等优点,成活率可达88%.结论:利用带节茎段能够实现铁皮石斛快速繁殖,为其人工扩大栽培奠定了良好基础.

Rapid propagation system for tissue culture of Dendrobium officinale Kimura et Migo

Abstract::Objective: In order to rationally protect and utilize the medicinal plant resources of Dendrobium officinale Kimura et Migo, plant tissue culture of stem - segment with node on Dendrobium officinale Kimura et Migo was studied. Methods : Using stem - segment with node of Dendrobium officinale Kimura et Migo as explants to analyse the effects of different culture medium and hor mones on induction, proliferation and rooting of Dendrobium offwinale Kimura et Migo. Results : The combination of 1.5 mg L- 1 6 - BA and 0.5 mg L-1 NAA added in the 1/2 MS medium(pH 5.6 5.8) was the optimal medium for buds induction with induc- tion rate of 93.3% ; the optimum medium for multiple shoots was the combination of 1.5 mg L-1 6 -BA and 0.5 mg L- NAA with 1/2 MS medium ( pH 5.6 - 5.8 ) and the average multiplication coefficient was 5.46 ; It was easy to rooting under the conditionof 0.5 mg L 1 IBA, 0.5 mg L-I NAA and 0.2% activated carbon (AC) with 1/2 MS medium ( pH 5.6 - 5.8) which made more roots and the rooting rate was 72%. Above all mediums' agar concentration were 0.8% and their sucrose concentration were 2%. At the same time, the fine woods were the best base material for seedlings with many advantages such as loose, breathable and good drainage and etc with survival rate of 88%. Conclusion: An effective approach for rapid propagation has been provided using stem - segment with node, which could promote artificial planting of Dendrobium officinale Kimura et Migo.
