4-1BB体外刺激的T淋巴细胞抑制小鼠黑色素瘤的作用

目的 探讨4—1BB体外刺激下小鼠T淋巴细胞的增殖情况及对小鼠黑色素瘤的抑瘤效果．方法：在IL-2＋CD3＋CD28基础上，增加4—1BB为刺激条件，体外培养经诱导的小鼠腹股沟淋巴结T细胞，MTT法检测其体外增殖率，并建立小鼠黑色素瘤实验模型，观察所诱导T淋巴细胞的抑瘤效果．结果：增加4—1BB后，T淋巴细胞的增殖率明显增加（1．15±0．08）％协（0．57±0．06）％，P〈0．01]，将所诱导的T淋巴细胞注入小鼠模型，其肺转移灶数目明显减少（93±6）vs（151±22），P〈0．01]，生存率提高，与对照组比较差异有统计学意义（P〈0．01）．结论：4—1BB在体外可促进T淋巴细胞的增殖，并具有较强的抗黑色素瘤作用．

Inhibition of mouse melanoma by 4-1BB-stimulated T lymphocytes in vitro

AIM: To analyze the proliferation of T lymphocytes in vitro stimulated by 4-1BB, and to observe the effects of 4-1BB-stimulated T lymphocytes on mouse melanoma model. METHODS: Besides IL-2+CD3+CD28, 4-1BB was added to stimulate T lymphocytes from mouse inguinal lymph node, and MTT was applied to test the proliferation rate. Mouse melanoma models were also set up, and the effects of stimulated T lymphocytes on tumor progression were also observed. RESULTS: Addition of 4-1BB greatly increased the proliferation of cultured T lymphocytes in vitro. After transferring of these cells to mouse melanoma models, the lung metastasis rate decreased, and the survival rate increased greatly, with a significant difference from the controls (P<0.01). CONCLUSION: 4-1BB can promote the proliferation of T lymphocytes in vitro, and has potent inhibitive effects on the melanoma.
