人脑星形胶质细胞瘤中nm23-H1基因和蛋白的表达

摘    要:目的:研究人脑星形胶质细胞瘤中nm23-H1基因及蛋白的表达,并初步探讨其临床意义。方法:采用地高辛标记的nm23-H1反应cRNA探针,通过原位杂交方法检测21例新鲜星形胶质细胞瘤标本中nm23-H1mRNA,并采用鼠抗人nm23-H1单克隆抗体,通过免疫组织化学的方法检测62例人脑星形胶质细胞瘤石蜡标本中nm23-H1蛋白。结果:21例新鲜标本的原位杂交结果显示低级别组星形胶质细胞瘤中nm23-H1mRNA的表达显高于高级别组。62例人脑星形胶质细胞瘤免疫组化结果,低级别组中nm23-H1蛋白的表达高于高级别组。nm23-H1mRNA原位杂交结果与nm23-H1蛋白免疫组化结果具有一致性。结论:nm23-H1mRNA及其蛋白低表达与星形胶质细胞瘤的高病理学分级有关,提示两再作为临床判断星形胶质细胞恶性生物学行为的参考指标。

Abstract:Objective: : To study the expression of nm23-H1 gene and pr otein in human astrocytoma and discuss its clinical significance. Method s: Expression of nm23-H1 mRNA was detected in 21 fresh specimens of ast rocytoma by in situ hybridization using nm23-H1 antisense complementary RN A probe. Expression of nm23-H1 protein was also detected in 62 cases of astrocy toma by immunohistochemical staining using monoclonal mouse anti-human nm23-H1 . Results: Result of in situ hybridization in 21 fresh speci mens: expression of nm23-H1 mRNA in the group of low grade astrocytoma was sign ificantly higher than that in group of high grade astrocytoma. Result of immunoh istochemical staining in 62 paraffin specimens: expression of nm23-H1 protien i n the group of low grade astrocytoma was higher than that in group of high grade astrocytoma. The expression of nm23-H1 mRNA were consistent with the expressio n of nm23-H1 protein. Conclusion: Low expression of nm23-H1 mR NA and protien is correlated with the high pathological grade of astrocytoma. It is implied that expression of nm23-H1 mRNA and protein can be used as the refe rence index to estimate the malignant biological behavior of astrocytoma.
