大黄素与黄芪甲甙对BXSB小鼠肾小球ICAM-1表达的影响

摘    要:目的 探讨大黄素与黄芪甲甙对BXSB狼疮样小鼠肾脏病变的影响及其免疫药理学机制。方法 将BXSB雄性小鼠 40只随机、均等分成 5组 (Ⅰ~Ⅴ组 )。Ⅰ组当即检测各项指标 ;Ⅱ~Ⅴ组分别胃饲生理盐水、大黄素、黄芪甲甙和两者混合物 ,40天后检测 2 4h尿蛋白浓度、血清抗核抗体 (ANA )滴度和肾脏细胞间黏附分子 1(ICAM 1)表达水平。结果 与Ⅰ组相比较 ,生理盐水组的各项指标都上升 ;与生理盐水组相比较 ,胃饲药物组的各项指标皆下降 ,其中以混合药物胃饲组下降最明显 ;肾小球ICAM 1表达与 2 4h尿蛋白浓度呈正相关。结论 大黄素和黄芪甲甙均对狼疮性肾炎有治疗作用且存在协同效应 ;抑制肾小球ICAM 1表达可能是其免疫药理学机制之一。

Abstract:Objective To study the effects of emodin and astragaloside on lupus nephritis (LN) and explore their possible immuno-pharmacological mechanism.Methods Forty male BXSB lupus like mice were randomizedly divided into five groups (GroupⅠ~Ⅴ).All indexes of GroupⅠ were detected at once. The other four groups were fed with simple normal saline (GroupⅡ), emodin(GroupⅢ),astragaloside (GroupⅣ) and the mixture of both emodin and astragaloside (GroupⅤ) respectively.The proteinuria concentration of 24 h, serumal ANA titers and ICAM 1 expression in glomeruli were detected after 40 days’ feeding.Results Compared with Group Ⅰ,all indexes of GroupⅡ were increased.Compared with GroupⅡ,the 24h proteinuria concentration,serumal ANA titers and ICAM 1 expression in glomeruli of GroupⅢ~Ⅴ were lower.These data of GroupⅤ were the lowest.The 24 h proteinuria concentration had a positive correlation with ICAM 1 expression.Conclusion Both emodin and astragaloside have therapeutic effects for LN.The possible immuno pharmacological mechanism may be depressing ICAM 1 expression in the glomeruli.
