人卵泡液及胚胎培养液中活性氧和抗氧化能力的变化研究

摘    要:目的研究超促排卵周期中不同直径卵泡液及不同质量胚胎培养液中活性氧(ROS)和抗氧化能力(TAC)含量的变化,探讨ROS和TAC在卵泡、胚胎发育中的作用及与妊娠结局的关系。方法采用比色法测定31例因输卵管因素行IVF-ET的患者不同直径卵泡液及不同级别胚胎培养液中的ROS、TAC含量。结果检测到大、中、小卵泡中均存在ROS、TAC,含量差异无统计学意义,但妊娠组TAC/ROS比值小于未妊娠组。在胚胎培养液中仅检测到ROS含量,未检测到TAC。优质胚胎与劣质胚胎间ROS含量差异无统计学意义,但当移植胚胎培养液中ROS〈200U/mL时,趋向有较好妊娠结局。结论 ROS和TAC与卵泡发育无关,但TAC/ROS比值增高可能与不良妊娠结局有关。胚胎培养液中ROS含量在不同级别胚胎差异无统计学意义,但培养液中ROS的低水平与良好妊娠结局有关,可能可以作为预测胚胎发育潜能的指标。

Abstract:Objective To study the changes of the reactive oxygen species(ROS)and total antioxidant capacity(TAC)in human follicular fluid and embryo culture media to search for the possible role in human follicular and embryo development.Methods Follicular fluid from 31 women undergoing IVF were divided into three groups via the diameter.And their embryo culture media on the third day after insemination were divided into two groups('good' or 'poor')according to a scoring system based on an assessment of the morphological appearance.ROS and TAC were determined by colorimetric method.Results The ROS and TAC levels in different diameter follicular fluid were no significant difference,and the ratio of TAC/ROS was less in pregnancy group.The ROS level can be tested in embryo culture media but TAC can not.The levels of ROS were no different in good and poor embryo culture media.There were no association between ROS level and IVF outcomes too.However,when ROS level less than 200U/ml in one of culture media,the pregnancy rates increased.Conclusion The study showed that ROS level in follicular fluid could not be a marker for predicting success in IVF patients.Even though there was no difference in good and poor embryo culture media,low levels in embryo culture media may be a marker for predicting success.
