雷公藤醇提物小鼠急性毒性实验

摘    要:目的对雷公藤醇提物进行急性毒性实验,了解雷公藤醇提物的急性毒性作用,初步判断可能的主要毒性靶器官。方法雷公藤醇提物设6个剂量组,给药剂量按生药折合计算,分别为10.0 g·kg-1、8.0 g·kg-1、6.4 g·kg-1、5.1 g·kg-1、4.1 g·kg-1、3.3 g·kg-1。口服灌胃一次给药,连续观察14 d,记录小鼠毒性反应情况。结果雷公藤10.0 g·kg-1组小鼠肝脏表面呈细颗粒状,有的小鼠肝脏局部变成灰白色,病理切片见10.0 g·kg-1组、8.0 g·kg-1组肝细胞肿胀、胞质内有大小不等的空泡以及数量不一的肝细胞凋亡,肝细胞间有大量红细胞;6.4 g·kg-1组、5.1 g·kg-1组可见肝细胞凋亡。半数致死量(LD50)为5.6 g·kg-1,95%的可信区间为4.8~6.4 g·kg-1。结论根据雷公藤醇提物急性毒性实验的主要表现和死亡小鼠的大体解剖可以初步判断雷公藤醇提物致死的主要原因为肝毒性。

Abstract:Objective To conduct the acute toxicity experiment of Tripterygium Wilfordii extract (TW extract)and understand the acute toxicity effect of TW extract so as to preliminarily judge the potential main toxicity target organs. Methods Six dose groups of TW extract were set up, as 10.0 g·kg-1,8.0 g·kg-1 ,6.4 g·kg-1 ,5.1 g·kg-1,4.1 g·kg-1 and 3.3 g·kg-1 according to equivalent crude drug calculation. The oral gastric infusion was applied and the observation lasted for 14 days. The toxic reaction was recorded in the mice. Results In 10.0 g·kg-1 group,the fine granular form was showed on liver surface and the liver changed as grey white in local area. In 10.0 g·kg-1 and 8.0 g·kg-1 groups,the pathological sections indicated liver cellular swelling, vacuole of different sizes in cytoplasm and liver apoptosis, as well as a large amount of red blood cells in liver cells. In 6.4 g·kg-1 and 5.1 g·kg-1 groups, liver apoptosis presented. The half lethal dose( LDs0 ) was 5.6g·kg-1 and 95% CI was 4.8 to 6.4 g·kg-1. Conclusion According to the main manifestation of the acute toxicity experiment of WT extract and the gross anatomy of dead mice, it is judged preliminarily that the main lethal reason of WT extract is liver toxicity.
