DNA修复基因hOGG1多态性与HIV-1易感性的关系研究

摘    要:目的探讨我国汉族人群中8-羟基鸟嘌呤糖苷酶1(hOGGl)基因Ser326Cys多态性与HIV感染间的关系。方法采用病例-对照的流行病学方法,运用PCR-限制性片段多态(restriction fragment length polymorphism,RFLP)技术,对126例HIV血清学阴性者和154例HIV血清学阳性者hOGG1基因第326位点Ser/Cys多态性进行分析,并比较hOGG1不同基因型与HIV感染间的关系。结果HIV血清学阴性组中3种基因型频率分布与现报道的中国汉族人群结果相近,其Ser/Ser、Ser/Cys和Cys/Cys基因型分布频率分别为20.6%,46.8%和32.5%,HIv血清学阳性组分别为16.2%,51.9%和31.8%,两组间差异无统计学意义(χ^2=1.12,P〉0.05)。HIV血清学阳性组人群中,合并HCV感染者与未合并HCV感染者Ser/Ser、Ser/Cys和Cys/Cys基因型频率分别为:HCV阳性组20.0%、48.9%和31.1%;HCV阴性组14.7%、53.2%和32.1%,两组间差异无统计学意义(χ^2=0.68,P〉0.05)。结论本研究未发现hOGG1基因Ser326Cys多态性与HIV感染及HIV与HCV共感染间的相关性。

Abstract:Objective To explore the relationship between the polymorphism of hOGG1 gene and infection susceptibility of HIV-1. Methods DNA from 126 HIV negative and 154 HIV positive subjects were extracted using Qiagen DNA Blood Mini Kit. The allelic frequency was identified by means of PCR-RFLP analysis. Allelic polymorphism in these two populations was analyzed by χ^2 test. Results The frequency of three alleles was as follows: Ser/Ser 20.6 %, Ser/Cys 46.8 % , and Cys/Cys 32.5 % in HIV-1 negative group, Set/Set 16.2% , Ser/Cys 51.9% , and Cys/Cys 31.8% in HIV-1 positive group. The frequency of three alleles was as follows : Ser/Ser 20.0 % , Ser/Cys 48.9 % , and Cys/Cys 31.1% in HCV positive and HIV-1 positive group, Ser/Ser 14.7%, Ser/Cys 53.2%, and Cys/Cys 32.1% in HIV-1 positive and HCV negative group. No statistical differences were found between the HIV-1 negative and positive group, HCV positive and HIV-I positive group and HIV-1 positive and HCV negative group. Conclusions The results suggest that it is not found the relationship between the polymorphism of hOGG1 Ser326Cys and the HIV infection and HIV/HCV co-infection.
