Abstract:Objective To investigate effects of gastrodin on the expression of H1,H2 and H3 subtypes of histamine receptor in the medial vestibular nucleus(MVN)in rats after unilateral labyrinthine(UL)damage and its role in vestibular compensation mechanisms.Methods Healthy SD rats(n=99)were randomly divided into a normal control group(n=9),a sham operation group(n=18)and a study group(n=72).In the study group,chloroform was injected into the left tympanum to destroy the labyrinth,while the same amount of normal saline was injected in the sham operation group.After modeling confirmation,rats in the study group were randomly selected to receive high,medium or low dose of intramuscularly injected gastrodin(once a day for 3 days)or no gastrodin(control).RQ-PCR,immunohistochemistry and Western-Blot were used to evaluate expression of histamine receptors H1,H2 and H3 mRNA and proteins in the MVN.Results Compared with the normal control group,expression of histamine receptors H1,H2 and H3 mRNA and proteins in the MVN in modeling animals was up-regulated(P<0.05),especially in those treated with high dose of gastrodin,which was higher than in those with no gastrodin treatment(P<0.05).Conclusion Gastrodin may play an important role in vestibular compensation by up regulating expression of histamine receptors H1,H2 and H3 mRNA and proteins in the MVN in the presence of vestibular dysfunction.
