左归丸对肾虚大鼠MSCs增殖的影响

目的探讨补肾益精代表方左归丸对肾虚大鼠骨髓间充质干细胞(MSCs)增殖的影响。方法 SD雌性大鼠双侧卵巢切除增龄3月法复制肾虚模型,用左归丸给予部分模型大鼠灌胃。分离、培养正常组、模型组、左归丸组大鼠MSCs。观察各组大鼠MSCs细胞形态,生长周期,P3、P5代细胞生长曲线,β-半乳糖苷酶衰老细胞染色结果。结果模型组大鼠P3、P5代MSCsOD值明显低于正常组、左归丸组(P<0.05)。各组大鼠MSCs细胞周期无明显差异(P>0.05)。模型组衰老细胞阳性率最高,明显高于正常组(P<0.01),但与左归丸组间差异不明显(P>0.05)。结论去卵巢大鼠MSCs增殖能力减弱,左归丸一定程度上改善肾虚大鼠MSCs增殖能力。

Effect of Zuogui Pill on MSC Proliferation in vitro

Objective To investigate the effect of Zuogui pill on bone marrow mesenchymal stem cell(MSC) proliferation in vitro.Methods The weak kidney animal model was established by ovariectomy on female Sprague-Dawley(SD) rats;some model animal were fed with Zuogui Pill for 3 months.MSCs was isolated and purified by whole bone marrow adherent method.MSC proliferation capacity of normal group,model group,and Zuogui Pill group was determined by observing morphologic characteristics,growth cycle,growth curve of P3、P5 MSCs and β-galactosidase staining senescent cells of those.Results From growth curve of P3,P5 MSCs,the proliferation capacity of MSCs in model group was significantly lower than those of normal group and Zuogui pill group(P<0.05);β-galactosidase staining showed that the model group had the highest positive rate of aging cells,significantly higher than normal group(P<0.01).Conclusion To some extent,the decreased proliferation capacity of MSCs from ovariectomized rats can be improved by Zuogui Pill.
