银杏叶提取物温度敏感型鼻用原位凝胶的研究

摘    要:目的研究银杏叶提取物温度敏感型鼻用原位凝胶的工艺制备、体外释放、药效学。方法以黏度为指标,用正交实验来确定最佳工艺;通过透析试验,研究凝胶释放机制;采用小鼠记忆获得、记忆巩固和记忆再现实验进行药效研究。结果通过正交实验,确定泊洛沙姆188-泊洛沙姆407为15∶20,壳聚糖0.4%,HP-β-环糊精30%为最佳制备工艺;通过体外释放研究,表明药物是通过扩散作用进入体内而不是通过凝胶溶蚀使药物释放;不同剂量银杏叶提取物原位凝胶改善小鼠学习记忆功能障碍。结论银杏叶提取物温度敏感型原位凝胶具有良好的缓释性能,通过鼻腔给药,可达到提高脑功能的作用。

Abstract:Objective To study the extract of Ginkgo biloba leaves in temperature-sensitive in situ gel for nasal use. Methods With viscosity as the index,an orthogonal experiment was conducted to determine the optimal process. The learning and memory abilities,learning-behavioral training,learning and memory-behavior of mice were observed to study pharmacodynamics. Results Orthogonal experiments showed that the best technology involved poloxamer 188 and poloxamer 407 at a ratio of 15∶ 20,Chitosan 0. 4%,and HP-β-CD 30%. In vitro release studies indicated that the drug spread into the body rather than through gelatin dissolution. Different doses of Ginkgo biloba extract in situ gel could alleviate learning and memory dysfunction. Conclusion Temperature-sensitive in situ gel of extracts of Ginkgo biloba leaves can increase the absorption of the drug and improve bioavailability.
