依达拉奉对沙土鼠缺血-再灌注损伤后的脑保护作用机制的探讨

摘    要:目的:探讨依达拉奉对沙土鼠全脑缺血再灌注后自由基损伤的脑保护机制。方法:实验动物随机分为正常对照组、假手术组、手术组、手术 依达拉奉组,手术 依达拉奉组给予腹腔注射依达拉奉10mg/kg,12h/次。设缺血再灌注6、12、24、36、48、60、72h7个时间点,分别检测脑组织中超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,并观察每个时间点电镜下海马CA1区锥体细胞超微结构。结果:每个时间点手术 依达拉奉组动物脑组织中SOD活力较手术组增强,MDA含量较手术组下降,且每个时间点两组间比较差异均有统计学意义(P<0.05)。电镜超微结构显示手术 依达拉奉组细胞病理改变较手术组轻,48h改变最明显。结论:脑缺血缺氧早期使用依达拉奉,可增强缺血脑组织SOD活力;降低MDA含量,从而达到清除氧自由基、抗脂质过氧化、减轻再灌注损伤、保护脑细胞的作用。

Abstract:Objective: To investigate the brain-protective mechanism of edaravone in gerbils with cerebral ischemia-reperfusion injury. Methods:Animals were randomly divided into 4 groups, namely control group , sham-operation group, operation group and operation plus edaravone group.The operation plus edaravone group was administered edaravone 10 mg/kg body, twice a day, by abdominal injection. The SOD activities and the MDA content in the cerebral tissue were detected at the time of 6,12,24,36,48,60, 72 h after reperfusion respectively, meanwhile the supermicro-structure of the neurons of the hippocampal CA1 territory was observed under the electron microscope at each time point. Results:After injection, the SOD activities in the cerebral tissue of the operation plus edaravone group at each time point increased, while the MDA content decreased significantly compared with the operation group(P < 0.05). The supermicro-structure under the electron microscope indicated the pathological changes of the operation edaravone group were slighter than the operation group,and most obvious at the time of 48h after reperfusion. Conclusion:The results suggest that the administration of edaravone at early stage of cerebral ischemia and hypoxia could increase the SOD activities and decrease the MDA content, then eliminating oxygen free radicals, inhibiting lipid peroxidation and reducing reperfusion injuries.
