载骨髓间充质干细胞明胶微球的制备

摘    要:目的研究制备携载大鼠骨髓间充质干细胞(Bone marrow mesenchymal stem cells,MSCs)明胶微球的方法,探寻骨组织工程的新路径。方法以改良的双相乳化法制备载MSCs的明胶微球并与京尼平分别进行12h,24h,48h的交联,与未交联组对比。检测微球的粒径、交联度、溶胀率、载细胞率,并通过荧光染料和扫描电镜检测其内细胞存活性。结果交联时间控制在24h内各组明胶微球的粒径变化不大,分别为(75±15)μm、(81.58±24.68)μm和(108.64±37.44)μm,24h后微球粒径明显减小为(27.88±7.46)μm,差异具有统计学意义(P<0.05);交联过的明胶微球降解时间明显延长,较未交联组差异具有统计学意义(P<0.05)。结论京尼平交联24h后的明胶微球粒径大小适中,降解时间以及载细胞率符合实验要求,微球内携载的MSCs存活良好。

Abstract:Objective To study the method to prepare gelatin microspheres carrying rat bone marrow mesenchymal stem cells(MSC).Methods Gelatin microspheres carrying rat bone marrow MSC were prepared using the modified emulsification technique,which were cross-linked to genipin for 12,24 and 48h respectively,and compared with those of bone marrow MSC uncross-linked to gelatin.The diameter,cross-linked degree,swelling rate,bone marrow MSC-carrying rate of gelatin microspheres were detected.Survival rate of gelatin microspheres-carrying bone marrow MSC was assayed by scanning electron microscopy with fluorescence staining.Results The diameter of gelatin microspheres did not change significantly within 24h cross-linking,which was(75±15)μm,(81.58±24.68)μm and(108.64±37.44)μm,respectively.However,the diameter of gelatin microspheres decreased to 27.88± 7.46μm after cross-linked for 48 hours(P<0.05).The degradation time of cross-linked gelatin microspheres was significantly longer than that of uncross-linked gelatin microspheres(P<0.05).Conclusion The diameter of gelatin microspheres is moderate 24h after cross-linked to genipin.The degradation time and bone marrow MSC carrying rate can meet the requirements of experiment.The gelatin microspheres-carrying MSC can survive quite well.
