大鼠妊娠期补钙对新生仔鼠长骨干骺端钙盐沉着及骨密度的影响

摘    要:目的:研究妊娠期母体补钙与新生儿长管状骨干骺端钙盐沉着的关系及 其与发育过程中骨密度之间的关系,为临床早期处理儿童缺钙引起的骨发育障碍提供依据。方法:取健康未曾妊娠过的Sprague Daxley(SD)雌鼠20只,将其与雄合笼交配,将妊娠的大鼠分三组:高钙组、低钙组和正常钙组,分别给予含钙1.4%、0.02%、0.6%的饲料,其它匀相同^[1],一直喂到仔鼠出生后的第21天。之后三组的所有仔鼠均给予政党钙饲料直至第63天,在此期分别于出生第1天、第21天、第42天和第63天,取部分仔鼠的股骨、胫骨的干骺端测其钙含量,并测第63天仔鼠股骨、胫骨的骨密度。结果:仔鼠出生第1天,高钙组干骺端钙含量较高,正正常组相比差异有显著性,低钙组与正常组相比差异无显著性,仔鼠出生后第21天,三组的干骺端钙盐含量均有所下降,第42天又有所上升,至第63天高钙组与政党组相比,干骺端钙含量较低,并差异有显著性,低钙组比正常组的干骺端钙含量高,并且差异也具有显著性。第63天股骨、胫骨骨密度的结果与第63天股骨胫骨,干骺端钙含量的结果相一致,低钙组骨密度比正常组高,差异显著;高钙组比正常组骨密度低,差异也具有显著性。结论:妊娠期补钙可以提高胎鼠长骨干骺端钙盐的沉着。妊娠期、哺乳期接受低钙的孕鼠,其后代的长骨干骺端钙盐沉着较高,且骨密度较高。妊娠期、哺乳期接受高钙的孕鼠,其后代长骨干骺端钙盐沉着较低,且骨密度较低。

Abstract:Objective To research on the relationship between maternal calcium supplement during pregnancy and neonatal calcium salt deposit in metaphysis of long bone shaft of neonatal pups as well as the bone density,and provide the basis for treating infantile osteodysplasia caused by calciprivia in early clinical stage.Methods 20 SD healthy virgin female rats were put in the same cages with male rats for copulation.Then the pregnant rats were divided into three groups including high calcium group,low calcium group and normal calcium group,which were give the forage of 1.4% Ca,0.02% and 0.6% Ca respectively and there was no difference in the other conditions of each group .The above forage had been fed until the 21st day after delivery.And all pups of the three groups were given normal calcium forage until the 63rd day ever since.During this period, metaphysis of femur and tibia of partial pups were extracted at the first day,the 21st day,the 42nd day and the 63rd day to measured the calcium content in meaphysis,In addition, the bone density of pups'femur and tibia were measured at the 63rd day.Results The calcium content in metaphysis in high calcium group was significantly higher than that in normal calcium group at the first day of pup's birth,and there was no significant difference between low calcium group and normal group.All the calcium salt content of metaphysis of the three groups declined at the 21st day of pup's birth and rose again at the 42nd day.However,at the 63rd day,the calcium content in metaphysis of high calcium group was significantly lower than that of the normal group.The low calcium group had higher calcium content of metaphysis and had significant differences when compared with the normal group.And the result of bone density in femur and tibia was coincident with the result of the calcium content of metaphysis in femur and tibia.The low calcium group had higher bone density than the normal group and had significant difference,while the high calcium group had lower bone density than the normal group and had significant difference as well.Conclusion Calcium supplement during pregnancy could promote the deposit of calcium slat in metaphysis of long bone.Different levels of calcium during pregnancy and lactation would produce the sequent effects on its descendants with normal dietary calcium from weaning to adults.If the maternal rats received low calcium during pregnancy and lactation,their descendants' calcium salt deposit of long bone metaphyisis and their bone density would became higher;and if the maternal rats received high calcium during pregnancy and lactation,their descendants would get the opposite result to the former.
