芳香化酶P450与环氧化酶-2在子宫内膜异位症和子宫腺肌病中的表达及意义

摘    要:目的研究细胞色素芳香化酶P450(P450arom)和环氧化酶-2(COX-2)在子宫内膜异位症(EMs)和子宫腺肌病(AM)在位和异位内膜中的表达及相关性。方法应用免疫组化Envision法和半定量逆转录聚合酶链反应(RT-PCR)技术检测正常子宫内膜、EMs及AM的在位和异位内膜中P450arom及COX-2的蛋白与mRNA的表达,并进行相关性分析。结果P450arom和COX-2在正常子宫内膜中无表达或弱表达,二者在EMs和AM在位和异位内膜中的表达均高于正常子宫内膜,差异均有显著性意义(P〈0.05)。P450arom与COX-2在EMs和AM异位内膜中的表达均高于在位内膜,差异均有显著性意义(P〈0.05)。P450arom在EMs和AM增殖期和分泌期表达无差异(P〉0.05)。COX-2在正常子宫内膜、EMs分泌期表达高于增殖期,差异均有显著性意义(P〈0.05),在AM表达不随月经周期变化。P450arom mRNA与COX-2mRNA在EMs中的表达水平呈正相关(r=0.964,P〈0.01);在AM中的表达水平亦呈正相关(r=0.813,P〈0.01)。结论P450arom和COX-2在EMs和AM中呈过表达,与EMs和AM的发病相关,二者协同作用促进EMs和AM的发生发展。

Expressions of aromatase cytochrome P450 and cyclooxygenase-2 in endometriosis and adenomyosis and significance

Abstract:Objective To investigate the expressions of aromatase cytochrome p450 and cyclooxygenase-2 (COX-2)in endometriosis (EMs)and adenomyosis (AM) and their correlations in EMs and AM. Methods Immunohistochemistry (IHC)and RT-PCR were performed to detect the expressions of protein and mRNA of P450arom and COX-2 in normal endometrium , eutopic and ectopic endometrium of EMs and AM. Results Pd50arom and COX-2 was not expressed or weakly expressed in normal endometrium,the expressions of them in eutopic and ectopic endometrium of EMs and AM were both higher than those in normal endometrium(P〈0.05). Furthermore the expressions of P450arom and COX-2 in ectopic endometrium were both higher than those in eutopic endometrium of EMs and AM(P〈0.05).The expression of P450arom in EMs and AM was not differential between proliferative and secretory phase (P〉0.05).While the expression of COX-2 in secretory phase was higher than that in proliferative phase of normal endometrium and EMs (P〈0.05); but there was no cyclic change throughout the menstrual cycle of COX-2 in AM .The expression level of P450arom mRNA and cox-2 mRNA was directly correlated in EMs and AM respectively; the correlation coefficient r was 0.964 and 0.813 respectively (P〈0.01). Conclusion P450arom and COX-2 are over expressed in Ems and AM,they have a correlation with the pathogenesy of EMs and AM,the two synergistic action promote the genesis and pathological progress of EMs ad AM.
