中脑多巴胺能前体细胞分化过程中胶质细胞相关基因的表达

目的 分析原代培养的近交系胎鼠腹侧中脑前体细胞(ventral mesencephalic precursors,VMP)在诱导分化过程中胶质细胞相关基因的表达情况.方法 原代培养近交系胎鼠VMP,体外增殖培养7 d后诱导分化7 d.在第7天、11天和14天抽提RNA,用Affymetrix Rat Expression Array 230 2.0芯片检测基因表达谱,分析差异基因.结果 与胶质细胞密切相关的GFAP、S100b、Gjal和Clu基因均明显上调.而GDNF家族基因无明显改变.结论 在体外VMP的诱导分化过程中,随着星形胶质细胞的增多,GFAP、S100b、Gjal和Clu基因表达均明显上调,值得进一步深入研究.

Expression of astrocyte related gene during the differentiation of mesencephalic dopaminergic precursor cells in vitro

Objective To investigate the expression of astrocyte-related gene during the differentiation of primary cultured ventral mesencephalic precursors（VMPs） from inbred rat fetuses in vitro.Methods Primary cultured VMPs cells were isolated from inbred rat fetuses.The cells were harvested and cultured for 7 days,and then induced for differentiation for 7 days.Total RNA of the cells was extracted on day 7,day 11 and day 14,then gene expression profiles were determined by Affymetrix Rat Expression Array 230 2.0 chips.Results The expressions of astrocyte related genes,such as GFAP gene,S100b gene,gap junction protein alpha1（Gja1） gene,Clu gene were all up-regulated during differentiation of VMPs,while the expressions of GDNF family members were not significantly changed.Conclusion During differentiation of VMPs into astrocytes,the expressions of GFAP,S100b,Gja1 and Clu were all up-regulated significantly,further investigation remains needed.
