FR/CL体外干预人胃癌细胞株SGC-7901及人肺癌细胞株A549生长

摘    要:目的 研究金荞麦提取物FR联合刺梨提取物CL对人胃癌细胞株SGC-7901及人肺癌细胞株A549增殖及凋亡的影响.方法 MTT法检测FR、CL对SGC-7901、A549细胞的增殖抑制率;流式细胞术分析早期凋亡细胞比; RT-PCR、Western blot分析Ki-67及Bax、Bcl-2表达的变化.结果 FR、CL单药均可明显抑制SGC-7901、A549细胞的生长,呈剂量依赖性;两药IC_(30) 浓度联合呈增效效应; IC_(30) 浓度FR、CL单药和1/2 IC_(30) (FR)+1/2 IC_(30) (CL)联合对SGC-7901细胞的抑制率分别为(33.89±0.22)%、(30.86±1.17)%和(44.25±0.50)%,对A549细胞的抑制率分别为(30.91±1.72)%、(29.07±1.41)%和(43.62±2.15)%.流式细胞术结果表明,对照组、FR组、CL组、联合组细胞早期凋亡比例依次增加,两单药组及联合组与对照组之间、联合组分别与两单药组之间的差异均具统计学意义(P<0.05);与单药组比较,联合组Ki-67、Bcl-2 mRNA/蛋白表达率有显著降低,而Bax mRNA/蛋白表达显著增高.结论 FR、CL均可抑制人胃癌细胞株SGC-7901细胞、人肺癌细胞株A549的增殖和诱导其凋亡,且联合应用存在增效效应.

Extracts of Fagopyrum cymosum and Rosa roxburghii Tratt inhibit proliferation and induce apoptosis of human gastric and pulmonary carcinoma cells
Abstract:Objective To investigate the effects of extract of Fagopyrum cymosum (FR) and Rosa roxburghii Tratt (CL) on the human gastric carcinoma SGC-7901 and pulmonary carcinoma A549 cell line. Methods Human gastric carcinoma SGC-7901 cells and pulmonary carcinoma A549 cells were exposed to FR and/or CL in vitro. The inhibitory effects were evaluated by MTT assay. Cell apoptosis was detected by flow cytometry. The expressions of Ki-67,Bax and Bcl-2 in mRNA and protein level were quantified by RT-PCR and Western blot. Results The growth inhibition of SGC-7901 and A549 was dose-dependent after exposure to the FR or CL alone. The inhibitory rates of FR at IC_(30) ,CL at IC_(30) and FR plus CL at 1/2 IC_(30) were (33.89±0.22)%,(30.86±1.17)% and(44.25±0.50)% for SGC-7901; and(30.91±1.72)%,(29.07±1.41)% and(43.62±2.15)% for A549,respectively. The cells apoptotic rates increased in order of control,FR,CL and FR plus CL group (P<0.05). Compared with the FR or CL,the expressions of Ki-67 and Bcl-2 mRNA/protein in FR plus CL group were decreased but the expression of Bax mRNA/protein was increased,both for SGC-7901 and A549 cells. Conclusion Both FR and CL have inhibitory and apoptosis-inducing effects on the human gastric carcinoma SGC-7901 and pulmonary carcinoma A549 cell line,and the combination of FR and CL shows a synergistic effect.
