Forskolin抗金黄地鼠视网膜节细胞凋亡作用与cAMP和JNK关系的研究

利用 Hoechst332 5 8荧光核染色和琼脂糖凝胶电泳、免疫印迹技术以及 Brown放射免疫法等实验手段 ,分别观察了forskolin对视网膜节细胞凋亡、视网膜细胞 phospho-JNK的表达以及 c AMP水平的影响。结果表明 :在近端切断金黄地鼠的视神经 ,视网膜 c AMP水平降低 ,phospho-JNK水平升高 ,视网膜节细胞发生凋亡 ;玻璃体内注射 forskolin可使 c AMP水平升高 ,phospho-JNK水平下降 ,凋亡的视网膜节细胞密度降低。提示 ,forskolin可能通过升高 c AMP水平而抑制 JNK的活性和产生抗凋亡的作用

THE STUDY OF THE RELATION BETWEEN THE ANTI-APOPTOSIS EFFECTS OF FORSKOLIN ONRETINAL GANGLION CELLS AND cAMP AS WELL AS JNK IN ADULT HAMSTERS

Hoechst 33258 fluorescent staining method, DNA electrophoresis in agrose, western blot analysis and Brown's radioimmunoassay were used respectively to observe the effects of forskolin on the apoptosis of retinal ganglion cells(RGCs), the expression of phospho JNK and the level of cAMP in the retina of adult hamsters. The results showed that the level of cAMP reduced, the expression of phospho JNK elevated and the apoptosis of RGCs occurred after optic nerve transection. While intravitreal injection of forskolin could elevate the level of cAMP, inhibit the expression of phospho JNK and reduce the density of apoptotic RGCs. These results indicated that forskolin might inhibit the activity of phospho JNK and the apoptosis of RGCs by elevating the level of cAMP in retina.
