电针对脑缺血大鼠神经黏蛋白mRNA表达与细胞超微结构的影响

摘    要:目的观察电针对易卒中型肾血管性高血压大鼠(stroke prone renovascular hypertensive rats,RHRSP)脑缺血-再灌注后不同时间点神经黏蛋白mRNA(neurocan-mRNA)表达、细胞超微结构的影响。方法用环形银夹使SD大鼠的双侧肾动脉狭窄,制成RHRSP,再用线栓法制成一侧大脑中动脉闭塞(middle cerebral artery occlu-sion,MCAO)模型。运用原位杂交和电镜等技术观察电针对脑缺血2h后再灌注1、7、14、30d大鼠脑内神经黏蛋白-mRNA表达与细胞超微结构的干预作用,并与对照组比较。结果电针组脑缺血-再灌注7、14、30d大鼠脑缺血区周围及海马区neurocan-mRNA表达均低于同期对照组,差异有统计学意义(P<0.05);电针组神经元、血管壁等细胞超微结构的损伤较对照各组减轻。结论电针对RHRSP脑缺血-再灌注损伤的保护作用,可能与其下调神经抑制因子neurocan-mRNA表达、减轻细胞超微结构损害等机制有关。

Abstract:Objective To investigate the effects of the electric acupuncture on the expression of neurocan-mRNA and ultramorphology of brain tissue following the cerebral ischemia and reperfusion in Stroke-prone-Renovascular-Hypertensive-Rats(RHRSP).Methods Stroke-prone-Renovascular-Hypertensive-Rats(RHRSP) models were established by bilaterally constricting the renal artery with sliver loop clips and the models of rat middle cerebral artery occlusion(MCAO) were created by occluding middle cerebral artery with suture in RHRSP for two hours.The effects of the electric acupuncture on the expression of neurocan-mRNA and brain ultramorphology in the rat cerebral tissue were detected using hybridization in situ and electron microscopy 1,7,14 and 30 days after onset of ischemia.Results The expression of neurocan-mRNA in the ischemic and hippocampus area at 7,14 and 30 days after onset of ischemia,were significantly lower in electric acupuncture group than in control group(P<0.05).Compared with controls,acupuncture treatment significantly improved ischemia induced ultramorphological changes in neuron and vessel wall.Conclusions The electric acupuncture protects against ischemic brain injury in RHRSP through downregulating the expression of neurocan-mRNA and improving ischemic brain ultramorphology.
