摘 要: | 目的:研究透骨草提取物对人卵巢SKOV-3细胞凋亡的影响及其机制。方法:吖啶橙染色法观察透骨草提取物对SKOV-3细胞形态的影响;免疫组化法检测透骨草提取物对SKOV一3细胞Caspas-es蛋白表达的影响。结果:37.8μg/ml透骨草提取物作用于SKOV-3细胞24h后,SKOV-3细胞形态不规则,细胞核固缩、呈月牙形紧贴在核膜周围。SKOV-3细胞Caspase-9、Caspase-3、Caspase-7蛋白表达明显低于与对照组(P〈0.05),表明透骨草提取物可下调SKOV-3细胞Caspase-9、Caspase-3、Caspase-7蛋白的表达。结论:透骨草提取物可以通过Caspases依赖途径诱导SKOV-3细胞凋亡。

Abstract: | Objective: To investigate the influences of Phryma Leptostachya L. var. asiatica Hara extracts on expressions of Caspase proteins of SKOV -3 cells and related molecular mechanism. Methods: The morph of SKOV - 3 cells was observed with acridine orange staining method. The expressions of Caspase - 9, Caspase - 3 and Caspase -7 proteins of SKOV -3 cells were detected with immunohistochemistry technique. Results: The morph of SKOV - 3 cells was unregular, karyopyenosis aggregated beside edge of SKOV - 3 cell mem- brane with crescent after SKOV -3 cells were treated with 37.81μg/ml Phryma Leptostachya L. var. asiatica Hara extracts. The expressions of Caspase - 9, Caspase - 3 and Caspase - 7 proteins of SKOV - 3 ceils treated with 37.8 p,g/ml Phryma Leptostachya L. var. asiatiea Hara extracts decreased than those in the control group ( P 〈 0. 05 ), indicating Phryma Leptostachya L. var. asiatiea Hara extracts could downregulate expressions of Caspase -9, Caspase -3 and Caspase -7 proteins of SKOV -3 cells. Conclusion: Phryma Leptostachya L. var. asiatiea Hara extracts could induce the apoptosis of SKOV -3 cells by Caspase proteins pathway. |
