摘    要:目的探讨哮喘小鼠A2B腺苷受体表达与气道炎症及气道重塑的关系及其机制。方法建立小鼠哮喘模型,采用双抗体夹心ELISA法测定小鼠血清中IL-8的水平,采用HE染色和免疫组化技术结合计算机病理图像分析系统测定小鼠气道形态学参数及支气管上皮细胞A2B腺苷受体的相对含量。结果哮喘组血清IL-8含量明显高于对照组(P0.05);哮喘组支气管壁厚度和平滑肌厚度均明显高于对照组(P均0.05);哮喘组A2B腺苷受体在支气管上皮细胞的表达量明显高于对照组(P0.05)。结论 A2B腺苷受体的过度表达可能在哮喘气道炎症及气道重塑发生机制中发挥重要作用;A2B腺苷受体可能通过促进IL-8释放来干预哮喘小鼠气道重塑。

Abstract:Objective It is to observe the expression of A2B adenosine receptor in lung tissue of asthmatic mice,and explore the role of A2B adenosine receptor in airway inflammation and remodeling.Methods BALB/C mice were sensitized and excited by ovalbumin(OVA) to establish asthmatic models.The level of IL-8 in blood serum was measured by ELISA.The changes of mice's airway wall morphologic parameter and the relative content of A2B adenosine receptor in bronchial epithelium were detecteded by both HE staining and a computer assisted image analysis system combined with immunohistochemical technique.Results The level of IL-8 in blood serum in asthmatic group was significantly higher than that in control group(P<0.05).The thickness of bronchus wall and smooth muscle in asthmatic group were both significantly higher than those in control group(Pall<0.05).The amount of A2B adenosine receptor expression in bronchial epithelium in asthmatic group were significantly higher than those in control group(P<0.05).Conclusion The excessive expressions of A2B adenosine receptor maybe play an important role in the pathogenesis of airway inflammation and remodeling in asthma.It is maybe through evoking IL-8 release that A2B adenosine receptor contributes to airway remodeling in asthma.
