姜黄素对急性肺栓塞大鼠细胞凋亡的影响

摘    要:目的观察姜黄素对急性肺栓塞(PTE)后细胞凋亡的影响,并探讨其可能机制。方法健康雄性SD大鼠40只,随机分为5组(n=8):假手术组、模型组及姜黄素低剂组、中剂组、高剂组(腹腔注射50、100、150mg/kg)。采用颈总静脉注射自体血栓的方法制备大鼠PTE模型。取肺组织,采用流式细胞仪检测细胞凋亡率,RT-PCR和Western blot方法检测Bcl-2、Bax、Caspase-3、Caspase-9 mRNA及蛋白的表达。结果与假手术组比较,模型组细胞凋亡率及Bax、Caspase-3、Caspase-9 mRNA和蛋白表达明显上升,Bcl-2表达明显下降(P<0.01);与模型组比较,姜黄素能明显降低细胞凋亡率及Bax、Caspase-3、Caspase-9表达,升高Bcl-2表达(P<0.05和P<0.01)。结论姜黄素能抑制PTE大鼠的肺组织细胞凋亡,与下调Bax、Caspase-3、Caspase-9表达,上调Bcl-2表达有关。

Abstract:Objective To evaluate the effects of curcumine on apoptosis induced by acute pulmonary thromboembo- lism (FIE) in rats and the molecular mechanism. Methods 40 male SD rats were randomly divided into sham-operation control group, model group and high, middle, low dose curcumine group (50,100,150 mg/kg, ip), each of 8. PTE model was established by intravenous injection of auto-blood clots. Lung tissue was taken. The percentage of apoptosis cells were detected by flow cytometry (FCM). The expression levels of Bcl-2,Bax,Caspase-3 and Caspase-9 were assayed by RT-PCR and West- ern blot. Results Compared with sham-operation control group, the percentage of apeptosis cells, the expression of Bax, Caspase-3, Caspase-9 mRNA increased, while Bcl-2 decreased in model group, and the differences were statistically significant (P 〈 O. O1 ). Compared with the model group, the percentage of apoptosis ceils and the expression of Bax, Caspase-3, Caspase- 9 were down-regulated markedly, while Bcl-2 were up-regulated in curcumine group, and the differences were statistically sig- nificant (P 〈 0.05 or P 〈 0.01 ). Conclusion Curcumine can inhibit apoptosis of the lung tissue in PTE rat by down-regulating the expression of Bax ,Caspase-3 ,Caspase-9 and up-regulating the expression of Bcl-2.
