摘    要:目的探讨有机磷杀虫剂毒死蜱(chlorpyrifos,CPF)长期染毒对大鼠空间记忆的影响及其可能机制。方法选择不引起大鼠出现全身系统毒性的CPF剂量(1,5,10 mg/kg)对大鼠连续染毒12周,利用Morris水迷宫评价大鼠的空间记忆功能,利用Western印迹检测CREB信号通路的蛋白表达。结果 CPF染毒大鼠到达平台的潜伏期呈剂量依赖性增加;随着染毒剂量的增加,大鼠海马CREB及ERKⅠ/Ⅱ磷酸化蛋白逐渐减少,而CaMKⅡ磷酸化蛋白、PKA蛋白表达没有明显改变。结论 CPF长期染毒能够诱发大鼠空间记忆的损伤,ERK/CREB信号通路的抑制可能是参与该变化的重要信号途径。

Abstract:Objective To evaluate the changes in spatial memory and CREB signal systems involved in the repeated ex- posure to chlorpyrifos (CPF) in rats. Methods The CPF (0,1.0, 5.0 or 10.0 mg/kg) was administered daily to male Wistar rats for 12 weeks. Morris water maze was used to evaluate the spatial memory function and the Western blot was used to analyze the protein expression. Results The CPF treated rats took more time to reach the platform in the water maze test. The phosphorylated protein expression of ERK I / II and CREB decreased. Conclusion Chronic exposure to CPF could result in spatial memory impairment. The inhibition of ERK/CREB signal system may be the important pathway to regulate the CPF caused spatial memory deficit.
