RP-HPLC内标法测定牡丹皮中丹皮酚的含量

 目的建立一个测定牡丹皮原药材中主要成分丹皮酚含量的新方法,在新方法中以肉桂酸为内标准确测定了牡丹皮药材中的丹皮酚。方法色谱柱采用ZORBAX SB-C₁₈(4.6 mm×250 mm,5μm)柱,流动相为甲醇-水-冰醋酸(50∶50∶0.5),流速1 mL·min^-1,检测波长为280 nm,柱温:室温。结果当丹皮酚的质量浓度在5.85～117.0·mg·L^-1时,其峰面积与质量浓度的线性关系良好(r=0.999 9);平均回收率101.0%,RSD=1.5%。结论方法准确,可靠,可作为牡丹皮原药材或含有牡丹皮的中成药建立对丹皮酚含量测定方法时应用。

Determination of Paeonol in Cortex moutan by RP-HPLC

OBJECTIVE To establish a new internal standard RP-HPLC method with cinnamic acid as internal standard for quantitation of paeonol in Cortex moutan.METHODS The determination by RP-HPLC was carried out using ZORBAX SB-C₁₈ column(4.6 mm×250 mm,5 μm) at room temperature,with mobile phase of methanol-water-acetic acid(50∶50∶0.5) at a flow rate of 1 mL·min^-1.The detection wavelength was set at 280 nm.RESULTS Excellent linear correlation was established with the range of 5.85～117 mg·L^-1(r=0.999 9).The average recovery of this method was 101.0%,and RSD was 1.5%.CONCLUSION The method can be applied in quantitive determination of Cortex moutan or preparations contained Cortex moutan with good accuracy and reliability.
