摘    要:为了研究人红白血病HEL细胞有自分泌血小板生成素(Tpo)的能力,采用细胞原位杂交检测mRNA,采用免疫荧光方法、生物素亲和素系统检测蛋白质,采用细胞传感器检测细胞电化学行为。结果表明:Mpl是细胞膜受体;HEL细胞内存在Tpo蛋白质;在培养基中加入rhTpo后HEL细胞增殖;可溶性受体融合蛋白可以阻断HEL细胞增殖,阻断后HEL细胞的电子传递数减少,但细胞数高于接种细胞数。实验结论提示Tpo可能是HEL细胞的自分泌因子。

Abstract:In order to identify Tpo as the autocrine growth factor of human erythroleukemic cell line (HEL cells). non-radioactive in situ hybridization was used to detect mRNAs of Tpo and c-mpl, indirect immunofluorescence and biotin-avidin system were used to detect Tpo and Mpl, a cytosensor was used to analyze the electronic voltammetric behavior (EVB) of HEL cells. Results indicated: Mpl as a membrane receptor on HEL cells, Tpo protein existing in HEL cells, rhTpo promoting proliferation of HEL cells and Mpl-IgG blocking the proliferation of HEL cells. After the Mpl-IgG blocking, EVB of HEL cells decreased, but there was still a discordance between cell count and EVB of HEL cells. It was concluded that Tpo may be a possible autocrine factor of HEL cells.
