KSC联合紫杉醇对人肝癌HepG-2细胞株的抑制作用研究

摘    要:目的:研究硒酸酯多糖(KSC)与紫杉醇(TAX)联合对肝癌细胞株HepG-2的生长抑制作用,判断联合用药的效果。方法:采用MTT法测定药物的体外杀伤作用,应用金氏公式进行联合用药分析。结果:KSC与TAX单用均可抑制HepG-2细胞的生长,且呈时间—剂量依赖性,二者合用后对HepG-2细胞的抑制作用显著增强。两药合用的中效浓度分别为31.684mg/L和0.272nmol/L,较两药单用时的中效浓度低(KSC单用为46.632mg/L,TAX单用为1.684nmol/L)。两药合用的绝大多数q值在0.882~0.989,为相加作用。结论:KSC与TAX联用可显著增强对HepG-2细胞的抑制作用,二者具有协同相加抗肿瘤作用。

Abstract:Objectlve :To study the interaction of kappa-selenocarrageenan (KSC)and paclitaxel (TAX)on human hepatocellular carcinoma HepG-2 in vitro in order to judge the effect of combination.Methods:The MTT was used to estimate the killing effect of drugs and the King's principle was used to analyze union of medication.Results:KSC and TAX could respectively inhibit HepG-2 cells proliferation in a time and concentration dependent patterns ,which was enhanced significantly as combined with each other.The medianeffect concentrations (IC50)of KSC and TAX in combination were 31.684mg/L and 0.272nmol/L,which was tess than that of single use of KSC (46.632mg/L)and TAX (1.684nmol/L).The q values of most combinations were from 0.882 to 0.989 and they acted in an additive pattern.Conclusion: KSC combined with TAX can significantly enhance the effects of growth inhibition and they play an additive antitumor effect in hepatocellular carcinoma.
