斑点免疫金染色法与免疫金银染色法诊断日本血吸虫病的比较

摘    要:目的建立快速、敏感、特异性强,操作简便和经济实用的诊断日本血吸虫病的免疫学检测方法一快速斑点免疫金染色法(R-Dot-IGS)。方法将日本血吸虫虫卵可溶性抗原点加于微孔滤膜,经预作用和封闭后,依次加入待检血清和金标记二抗,然后观察结果,并与快速微量斑点免疫金银染色法(RM-Dot-TGSS)和斑点免疫金银染色法(Dot-IGSS)进行比较。结果用R-Dot-IGS、RM-Dot-IGSS和Dot-IGSS同步检测55份日本血吸虫病患者血清,三种方法的阳性率分别为98.18%(54/55)、98.18%(54/55)和100%;检测50例健康人血清均为阴性;检测10例肝吸虫病人和10例肺吸虫病人血清,仅有1份肝吸虫病人血清在RM-Dot-IGSS反应为阳性。结论R-Dot-IGS检测过程仅为15min不需银显影,具有敏感特异、快速简便实用的特点,适用于日本血吸虫病的现场流行病学调查和临床快速诊断。

Abstract:Objective To set up a fast, specific, sensitive, convenient and economical assay, rapid-dot-immunogold staining (R-Dot-IGS) for detecting serum antibodies of the patients infected with Schitosoma japonicum. Methods: The soluble egg antigen of Schitosoma japoncum were added onto microspore membrane. After pre-reacting and blocking, the serum to be detected and sheep anti-human IgG labeled with chloroauric acid were added sequentially to read the results. The assay only took about 15 min. As Controls, the Dot- immunogold sliver staining (Dot-IGSS) and rapid micro volume Dot-IGSS (RM-Dot-IGSS) were performed simultaneously. Results The positive rates were 98.18%, 98.18% and 100% respectively, when the serum of schistosomiasis japonica patients detected by the R-Dot-IGS, Dot-IGSS and RM-Dot-IGSS. The sera of 50 health controls, 10 clonorchiasis cases and 10 paragonimiasis cases all showed negative reactions, except one clonorchiasis cases that showed positive reaction with RM-Dot-IGSS. Conclusion Compared with Dot-IGSS and RM-Dot-IGSS, R-Dot-IGS has similar sensitivity and specificity, but the latter is more rapid, simple and more economical, and hence is worth for application in field situations and clinical rapid diagnosis of Schistosomiasis japonica.
