裸鼠盲肠黏膜下注射HCT116细胞建立结肠癌原位种植及转移模型的研究

目的建立一种简便、可行、稳定的裸鼠盲肠黏膜原位种植癌及转移模型。方法将人结肠癌细胞株HCT116细胞注射接种于裸鼠盲肠黏膜下,HE染色及人癌胚抗原(CEA)免疫组化检测接种后不同时期的盲肠原位瘤,腹腔淋巴结、肝、肺组织转移情况。结果盲肠接种HCT116细胞后2周,90%裸鼠均长出直径2~5 mm的盲肠原位瘤;接种后4周开始有腹腔淋巴结转移,12周时淋巴结转移率100%;接种10周时见肝、肺有转移,12周时肝、肺转移率分别为60%、39%。结论利用盲肠黏膜下原位接种HCT116细胞法建立裸鼠结肠癌模型,可模拟结肠癌临床原位发生、发展演变过程,方法简便且成瘤率高,为深入研究结肠癌转移机制的基础研究提供理想、稳定的动物模型。

Establishing orthotopic and metastatic colon cancer model by submucous injection of HCT116 cells in nude mice

Objective To establish a simple,feasible and reliable orthotopic implantation and metastasis model of human colon cancer in nude mice cecum mucosa.Methods Tumor cell line HCT116,derived from human colon cancer,was injected into colon cecum mucosa of nude mice to develop colorectal cancer model.HE staining and human carcinoma antigen (CEA) immunohistochemistry were performed to detect tumor in situ and metastasis condition in abdominal lymph nodes,the liver and the lungs.Results On the 2nd week after inoculation,90％ of the nude mice developed cecum tumor of 2-5 mm in diameter in situ.Abdominal lymph node metastasis was observed on the 4th week and present in all mice at week 12.Liver and lung metastasis started to appear at a percentage of 60％ on the 10th week and 39％ on the 12th week.Conclusion Human colon cancer in situ could be successfully established by inoculating HCT116 into cecum mucosa of nude mice to mimic the clinical development and growth of colon cancer.This method proves to be simple with a high tumor-bearing rate.It could be used as a reliable and ootimal animal model for in-depth basic research about the mechanism of colon cancer metastasis.
