慢性心力衰竭患者外周血单核细胞TNF-α mRNA转录和蛋白质表达的改变及开博通的干预作用

摘    要:目的 通过测定慢性充血性心力衰竭 (CHF)患者外周血单核细胞肿瘤坏死因子 - α(TNF- α) m RNA转录和蛋白质表达的改变 ,及血管紧张素转换酶抑制剂开博通对其的影响 ,探讨 TNF- α在 CHF发病中的作用、可能来源及与肾素 -血管紧张素系统的关系。方法 收集 2 0例 CHF患者及 10例健康人外周血单核细胞 (PBMC) ,应用 RPMI16 4 0培养基进行培养 ,并加入开博通 ,使开博通终浓度分别为 0 m mol/ L ,10 - 8mmol/ L。 2 4 h后 ,取培养 PBMC,应用 RT- PCR和Western blot技术测定 CHF患者和正常对照组外周血单核细胞 TNF-α m RNA转录和蛋白质表达的变化。结果 与正常对照组相比 ,CHF患者外周血单核细胞 TNF-α m RNA转录水平、TNF-α蛋白的相对含量高于对照组。开博通对CHF组 PBMC TNF- α m RNA转录和蛋白质表达有抑制作用。结论  (1) CHF患者 TNF- α m RNA及蛋白质表达增加 ,提示 TNF- α可能参与了心力衰竭的发生发展过程。 PBMC可能是心力衰竭时 TNF- α表达增加的一个来源。 (2 )TNF- α m RNA及蛋白质表达增加可能与心力衰竭时肾素 -血管紧张素系统被激活有关

Abstract:Objective To study the expression changes of tumor necrosis factor(TNF-α) mRNA and protein in patients with chronic heart failure and influence of angiotensin converting enzyme inhibitors(ACEI) on it. Methods Peripheral blood mononuclear cells (PBMC) was isolated from plasma, which was cultured with RPMI1640 medium for 24h,the conclusion of ACEI was respectively 0 mmol/L, 10 -8 mmol/L,then levels of TNF-α mRNA and protein of PBMC was determined by RT-PCR and Western blot in 20 patients with CHF and 10 health controls. Results The level of TNF-α mRNA and protein of PBMC of patients were higher than the controls. Capoten can inhibit the expression of TNF-α. Conclusion The expression of tumor necrosis factor in patients with chronic heart failure was up regulated, and ACEI can inhibit the expression of TNF-α,which suggested TNF-α and RAS interacted in the development of CHF and PBMC may be the one of source of TNF-α.
