摘    要:目的:研究古典型鼠疫菌与假结核菌ATCC29833基因组间的差异,为进一步认识鼠疫菌的起源与进化奠定基础。方法:通过抑制消减杂交技术比较两种细菌基因组间的差异。结果:与已测序的3株鼠疫菌基因组进行同源性比较,发现了259个假结核菌基因组中存在的特异序列,与基因库进行同源性比较发现了10个假结核菌ATCC29833基因组中存在的新序列。结论:抑制消减杂交技术是一个简单而有效的比较近源微生物基因组之间差异的方法,鼠疫菌在进化过程中失去了大量的基因,假结核菌基因组之间也存在着一定的差别。

Abstract:Objective: To reveal the difference between the genome of Yersinia pestis and that of Y. pseudotuberculosis for providing insight into the origin and evolution of Y. pestis. Methods: Genomic comparison between Y. pestis and Y. pseudotuberculosis was conducted by suppression subtractive hybridization(SSH). Results: A total of 259 fragments specific to Y. pseudotuberculosis ATCC 29833 were found based on sequence homologous analysis against three whole-genome sequences of Y. pestis, and 10 new fragments unique to Y. pseudotuberculosis ATCC 29833 were found based on sequence homology analysis against GenBank database. Conclusions: SSH technique is a simple and efficient tool for identifying genetic differences between the closely related strains of bacteria. A number of genes are deleted from Y. pseudotuberculosis during its evolution to Y. pestis, and Y. pseudotuberculosis has undergone genomic flux to some extent.
