人脑梗死后半暗带区细胞骨架与细胞凋亡的相关性研究

摘    要:目的:探讨人脑梗死后半暗带区细胞骨架与细胞凋亡的相关性。方法:应用10例脑梗死后的尸检脑标本,常规HE染色和免疫组织化学染色检测微管相关蛋白(MAP-2)、胶质纤维酸性蛋白(GFAP)、凋亡相关蛋白(Bcl-2),进行定性和定量分析。结果:脑梗死后以梗死灶为中心,按照组织损伤程度由内到外分为4个区(0-3区),其中2区为半暗带区;MAP-2在0区表达消失,1区和2区表达明显增强,3区表达正常;GFAP在0区和1区表达很少,2区和3区随缺血时间延长表达迅速持续升高;B -2在0区和1区表达很少,2区和3区表达显。结论:人脑梗死后涉及细胞骨架改变和细胞凋亡的发生,两呈正相关。对此过程中涉及到的因素进行干预,可能改变脑卒中的进程和预后。

Abstract:To investigate the relationship to cytoskeleton and apoptosis in ischemic penum-bra of human infarcted brains. Methods We used HE and immunohistochemical method to define changes of cells in post - mortern specimens from ten patients. Microtubule - associated protein 2 (MAP- 2) , glial fibril-lary acidic protein (GFAP) and anti - apoptotic protein Bel - 2 were stained as markers. Results Each sec-tion of the infarcted brain was divided into four areas (the area 0 - 3), and the area 2 was known as the ischemic penumbra. Immunoreactivity for MAP - 2 was disappeared in the area 0,increased in the area l and 2 apparently, and preserved in the area 3. The expression of both GFAP and Bel - 2 was lost in the area o and l, but preserved in the area 2 and 3. GFAP was selectively increased in the area 2 and 3,which manifest a time -dependent patten. Conclusions Changes of cytoskeleton and apoptosis that occur after ischemia may have a positive relationship with each other.
