摘    要:目的:制备拟Alzheimer病的简易大鼠模型。方法:大鼠海马CAI区微量注射冈田酸(Okadaic acid OA)隔日1次,共4次,采用Morris水迷宫测试大鼠空间学习记忆能力,免疫组化法观察大鼠海马过度磷酸化tau蛋白的表达,并与正常对照组比较。结果:与对照组比较,模型组大鼠学习记忆能力下降,海马磷酸化tau蛋白表达显著增强。结论:大鼠海马CA1区多次微量注射OA可制备简易拟Alzheimer病大鼠模型。

Abstract:Objective:To prepare a simple rat model of Alzheimer' s disease. Methods: Microinjection of Okadaic acid was given into the rat hippocampus CA1 region once every other day for 4 times totally. The spatial learning memory ability of rats was detected by Morris water maze. The expression of tau hyperphosphorylation was observed by immuno- histochemistry staining and compared with a normal control group. Results:Compared with the control group, the learning memory ability of rats in the model group was decreased significantly but the expression of protein-tau hyperphospho- rylation was obviously increased in hippocampus. Conclusion :That the microdosis of Okadaic acid is injected into hipp-ocamal CAI region of rat for many times might prepare a simple rat model of Alzheimer' s disease.
