玉屏风散提取液对小鼠腹腔巨噬细胞活化及增殖的影响

目的研究玉屏风散提取液对小鼠腹腔巨噬细胞增殖及活化的影响。方法分别制备玉屏风散水煎液、玉屏风散蒸馏水超声提取液、玉屏风散无水乙醇超声提取液和玉屏风散乙酸乙酯超声提取液。分离健康昆明种小鼠原代腹腔巨噬细胞进行体外培养,培养基中分别加入不同剂量上述药液使终末浓度达到1.67、3.33、8.33、16.67 g·L~(-1)(折合生药)进行细胞培养,以0 g·L~(-1)作为空白对照组。培养24、72 h后,分别加入中性红、2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐(WST-8),用酶标法检测4种玉屏风散提取液对腹腔巨噬细胞活化及增殖的影响。结果与空白对照组相比,4种不同浓度的玉屏风散提取液均可促进小鼠腹腔巨噬细胞吞噬中性红能力及增殖(P<0.01)。与同剂量玉屏风散水煎液相比,除玉屏风散无水乙醇超声提取液(3.33 g·L~(-1))、玉屏风散乙酸乙酯超声提取液(3.33 g·L~(-1))可促进巨噬细胞吞噬中性红能力(P<0.05,P<0.01),玉屏风散乙酸乙酯超声提取液(16.67 g·L~(-1))可促进腹腔巨噬细胞增殖(P<0.01),其余剂量的玉屏风散超声提取液的效果都不如同剂量组的玉屏风散水煎液。结论玉屏风散提取液可促进小鼠腹腔巨噬细胞活化吞噬能力及增殖,但玉屏风散的超声提取条件需进一步探索。
