摘    要:目的:探讨波棱瓜子脂肪油(HO)抗卡介苗(BCG)+脂多糖(LPS)诱导免疫性肝损伤的药理作用与其化学成分之间的关系,揭示其抗肝损伤的有效物质基础。方法:采用GC-MS建立9批HO的GC指纹图谱,并对各个峰进行标定,获得HO化学成分数据;采用BCG+LPS诱导造成免疫性肝损伤小鼠模型,分为空白组、模型组及9个给药组,给药组ig给予HO(7mL·kg-1)连续12 d,测定小鼠血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、超氧化物歧化酶(SOD)的活性,获得HO药理活性数据,将两者进行逐步多元回归分析,揭示HO抗肝损伤的有效物质基础。结果:建立了波棱瓜子脂肪油气相指纹图谱,并标定7个色谱峰;抗免疫性肝损伤药效实验中9个给药组均有不同程度的抗肝损伤作用,逐步回归分析表明HO抗免疫性肝损伤药理活性与其亚油酸、油酸、亚麻酸的含量成正相关。结论:HO对BCG+LPS诱导小鼠免疫性肝损伤的保护作用,可能与其含有不饱和脂肪酸有关,增强小鼠抗氧化活性。

Abstract:Objective:To perform the correlation between fingerprint and liver protection of Herpetospermum caudigerum seed oil (HO) illuminate the therapeutic basis of liver protection. Method:GC Fingerprint of HO was developed, and the protective effects of HO on immunologic liver injury model was observed,the contents of alanine aminotransferase(ALT),aspartate aminotransferase(AST) and superoxide dismutase(SOD) in the blood serum were measured.The correlation analysis between the GC fingerprint of HO and the contents of ALT,AST and SOD were carried out with stepwise regression. Result:The linoleic acid,oleic acid and linolenic acid were important for liver protection. Conclusion:The protective effects of HO on immunologic liver injury model might be related to its unsaturated fatty acids.
