灯盏花中咖啡酰化合物对溶血磷脂酰胆碱致牛脑微血管内皮细胞损伤的保护作用

摘    要:目的:研究灯盏花中3种咖啡酰化合物(DC-1,DC-2,DC-3)对溶血磷脂酰胆碱(LPC)引起的牛脑微血管内皮细胞(BCMEC)损伤的保护作用及抗氧化和抗活性氧作用。方法:方法:以乳酸脱氢酶(LDH)释放量为指标测定BCMEC损伤,比色法测定药物体外抗氧化和抗活性氧能力。结果:LPC(2.5μg/ml)gn BCMEC孵育24h后,可显增加LDH释放;3种咖啡酰化合物均可显抑制LDH释放,其作用强度DC3>DC2>DC1;且具有显的抗氧化和抗活性氧作用,其强度DC2>DC3>DC1。结论:灯盏花中3种咖啡酰化合物对LPC引起的BCMEC损伤具有明显的保护作用,可能与其抗氧化和抗活性氧作用有关。

Abstract:Objective: To study the protective effects of 3 caffeoylconjugation compounds from Erigeron breviscapus (DC-1,DC-2,DC-3) on lysophosphatidylcholine(LPC) induced damages of bovine cerebral microvascular endothelial cells (BCMEC) and their anti-oxidation and scavenging oxidation free radical ability. Methods: The injury of BCMEC was determined by measuring release of lactic dehydrogenase (LDH). The anti-oxidation and scavenging oxidation free radical effects were measured by colorimetric method. Results: When BCMEC was incubated with LPC(2.5 μg/ml) for 24 h, LDH release was markedly increased. Three caffeoyl conjugation compounds significantly inhibited LDH release and the strength order of the 3 compounds was DC3>DC2>DC1. They also possessed the ability of anti-oxidation and scavenging oxidation free radical and the strength order was DC2>DC3>DC1. Conclusion: Three caffeoyl conjugation compounds from Erigeron breviscapus protect BCMEC injury from LPC and the protective effects may be due to anti-oxidation and scavenging oxidation free radical.
