摘    要:目的探讨吲哚-3-甲醇(I3C)抑制人肝癌细胞株SMMC-7721生长作用及机理。方法采用人肝癌细胞株SMMC-7721制备荷瘤裸鼠;通过测量肿瘤大小和病理学检测,观察I3C对肿瘤细胞的抑制作用;采用流式细胞仪检测肿瘤细胞周期、线粒体膜电位;采用Western blot法检测相关蛋白的表达。结果经I3C作用后,SMMC-7721细胞生长变慢,细胞周期、线粒体膜电位、相关蛋白的表达改变。结论 I3C能够抑制SMMC-7721细胞生长,细胞周期和细胞凋亡相关信号分子可能在其中发挥了一定作用。

Abstract:Objective To study the effects and mechanism of Indole-3-carbinol on the inhibition of human liver cancer cell line SMMC-7721.Methods The tumor-bearing nude mice were prepared with human liver cancer cell line SMMC-7721 and observe the effects of inhibition by measuring the size and detection of pathological change of the tumor.Cell cycle and Mitochondrial membrane potential were detected with Flow cytometry.The expressions of the related protein were detected with western immublotting assay.Results I3C could inhibit the growth of SMMC-7721.The Cell cycle,Mitochondrial membrane potential and Protein expression changed.Conclusion I3C significantly inhibited the human liver cancer cell proliferation,which was probably related to some signal molecules about cell cycle and apoptosis.
