食管鳞癌的P53表达与DNA倍体关系

摘    要:目的;了解P53蛋白在食管鳞癌发生的作用。方法:应用Ab-6(DO-1)单抗对110例手术标本的石蜡切片进行自动免疫组化染色。结果:发现65%的食管鳞癌以及53%的癌旁正常上皮和84%的非典型增生一原位癌病变呈现P53蛋白过度表达。流式细胞分析显示79%为非整倍体肿瘤,21%的二倍体肿瘤,非整倍体肿瘤的P53阳性率明显高于二倍体肿瘤。结论:P53基因改变可能是食管鳞癌发生过程中的一个早期的生物学标

Abstract:Purpose To study the significance of p53 expression in the histogenesis of esophageal squamous cell carcinoma. Methods Accumulation of the p53 protein was examined immunohistochemically using monoclonal antibody Ab 6 in 110 routinely paraffin embedded esophageal squamous cell carcinomas specimens. Results p53 overexpression was present in 71 (65%) of 110 specimens. Positive p53 staining was also demonstrated in the foci of dysplasia and/or carcinoma in situ (21 of 25, 84%) and normal epithelia (38 of 72, 53%). DNA aneuploid was found in 79% (69 of 87) of the tumors by flow cytometry. The incidence of positive p53 staining was significantly higher in aneuploid tumor (74%) than that in the diploid tumor (50%) ( P <0 05). Conclusions These results suggest that p53 protein accumulation may be an early biomarker in the multistep process of esophageal carcinogenesis and that p53 expression is closely associated with cellular DNA ploidy.
