丙型肝炎患者血清中HCV F抗体的检测

目的以丙型肝炎病毒(hepatitisCvirus,HCV)F蛋白作为抗原,检测HCV感染者血清中的相应抗体,观察F抗体在国内HCV感染人群中的阳性率及其与HCV感染的相关性。方法基因工程重组表达的F蛋白经亲和柱纯化后,包被微量滴定板,以ELISA法检测35份HCV感染者、8份非HCV感染的肝病患者及10份健康者的血清标本。结合患者疾病资料,统计学分析F抗体与HCV感染的相关性。结果在35份HCV感染者血清标本中,F抗体的阳性率为43%(15/35),对照组(8份非HCV肝病患者及10份健康者)血清中无一例F抗体阳性,两组之间有显著的统计学差异(P<0.005)。但未发现F抗体阳性与患者年龄、性别、病程及血清HCV载量之间的相关性。结论本研究首次报道了F抗体在国内HCV感染人群中的检出情况,将有助于F蛋白的致病性研究,也对探讨F蛋白的功能及进一步将其作为HCV感染检测指标的研究有促进作用。

Detection of Serum Anti-F in Hepatitis C Virus Infected Patients

Objective To assess the prevalence of serum anti-F in patients with hepatitis C virus (HCV) infection, and the correlation between anti-F antibody and HCV infection. Methods Recombinant truncated HCV F protein was used as capture antigen to detect serum anti-F in blood samples collected from 35 patients with HCV infection,8 patients with other liver diseases, and 10 healthy individuals by enzyme-linked immunosorbent assay (ELISA). Results Fifteen samples (43%) out of the 35 HCV infected patients exhibited a positive anti-F reaction, showing significant difference from the controls without HCV infection (P<0.005). Chi-square test showed that there were no significant correlations between anti-F antibody and clinical status of the patients infected with HCV, including age, sex, stage of liver disease and HCV RNA serum level. Conclusion This first report of the serum anti-F prevalence in Chinese hepatitis C patients may help the investigation of the pathogenesis of HCV, and functions of F protein. The F protein can be used for diagnosis of HCV infection.
