摘    要:目的观察大豆异黄酮和叶酸对同型半胱氨酸糖尿病小鼠模型心肌组织血小板衍生生长因子-β(PDGF-β)表达的影响。方法昆明小鼠32只随机分4组,为正常对照组(NS组)、高同型半胱氨酸糖尿病组(HCYDM组)、大豆异黄酮组(SIF组)和大豆异黄酮加叶酸处理组(SIF+FA组),每组8只。各实验组HCY糖尿病小鼠模型通过尾静脉注射链脲佐菌素和腹腔注射同型半胱氨酸建立。检测血糖水平及PDGF-β的表达结果。结果(1)与NS组比较,各实验组血糖水平均升高(P<0.01);与HCYDM组相比较,SIF组及SIF+FA组血糖降低显著(P<0.05,P<0.01)。(2)与NS组相比较,HCYDM组心肌肥大,心肌细胞面积及核面积增大,PDGF-β在心肌细胞表达的平均光密度出现增高(P<0.01);与HCYDM组比较,SIF组与SIFFA组细胞面积减小(P<0.05,P<0.01)、核面积减小(P<0.01),PDGF-β表达的平均光密度降低(P<0.01)。结论大豆异黄酮和叶酸可抵抗高同型半胱氨酸糖尿病小鼠的心肌组织损伤,在糖尿病的防治上可能具有重要的临床意义。

Effect of soybean isoflavone and folic acid on the expression of platelet-derived growth factor-β in myocardial tissue of murine models with homocysteine diabetes

Abstract:Objective To observe the effects of soybean isoflavone and folic acid on the expression of platelet-derived growth factor-β(PDGF-β) in myocardial tissue of murine models with homocysteine (HCY) diabetes. Methods Thirty Kunming mice were divided randomly into 4 groups,which included normal control group (NS group), homocysteine diabetes group (HCYDM group), soybean isoflavone group (SIF group), and soybean isoflavone and folaein group (SIF + FA group), eight mice in each group. HCY diabetes models were made through vena caudalis injection of streptozocin and intraperitoneal injection of HCY. The blood glucose and the expression of PDGF-β were detected. Results ( 1 ) Compared with the NS group,the blood sugar level was higher in experimental groups( P 〈 0.01 ). Compared with the HCYDM group ,the blood glucose of the SIF group and the SIF + FA group were significantly lower( P 〈 0.05, P 〈 0.01 ). (2) Compared with the NS group, the HCYDM group showed cardiac muscular hypertrophia, and the myocardial cell area and nucleus area increased. Average optical of PDGF-β expressed in the myocardial cells rised up( P 〈 0.01 ). Compared with the HCYDM group, the cell area of the SIF group and the SIF + FA group decreased( P 〈 0.05, P 〈 0.01 ), the nuclear areas of the two groups also decreased( P 〈 0.0t ), Average optical of the expression of PDGF-β decreased ( P 〈 0.01 ). Conclusion Soybean isoflavone and folic acid are protecting factors which protect the myocardial tissue from damage in HCY diabetes murine model, and there are congenerous effects. They may play an important role in clinical treatment of diabetes.
