GR基因片段的克隆和可诱导的目标载体的构建

摘    要:为了构建针对小鼠糖皮质激素受体基因第2 外显子的可诱导目标载体,我们对小鼠糖皮质激素受体基因第2 外显子及其侧翼区域的基因组DNA 片段进行了结构分析,和部分片段的亚克隆,然后以pF1ox 质粒为框架构建了可诱导的目标载体。这一载体的构建为进一步建立诱导型GR 基因剔除小鼠奠定了基础,而且对于深入研究GR 基因的表达调控也很有帮助。

Abstract:To construct the inducible targeting vector for the mutagenesis of the mouse glicocorticoid receptor(GR) gene, we cloned and analyzed a genomic fragment containing the exon 2 and its flanking regions of the GR gene, and then, we subcloned some fine regions of the fragment. Basing on the feature of these regions and the requirement of knockout identification, we construct the targeting vector (pFloxGR), which provides the important base for the generation of the inducible knockout mice of the GR gene and its functional analysis.\=
