摘    要:目的采用HPLC法测定市售山楂中异槲皮苷及金丝桃苷的含量以建立质量控制方法。方法采用Ultimate@XB-C18色谱柱,采用梯度洗脱条件,检测波长:360 nm,流速:1 mL/min,柱温:30℃,进样量:10μL,对山楂中异槲皮苷、金丝桃苷进行检测,并进行系统适用性考察。结果经方法学验证,线性范围分别为0.60~38.52μg/m L、0.60~38.56μg/m L,相关系数均高于0.999;精密度、重复性的RSD均小于2%;平均回收率101.88%~102.44%,RSD小于2.5%。结论本法快速准确,可用于市售山楂异槲皮苷和金丝桃苷的含量测定,为其质量控制提供试验依据。

Abstract:Objective In order to establish the quality control method, isoquercitrin and hyperoside content of commercially available hawthorn were determined by HPLC. Methods When the Ultimate@XB-C18 column with different mobile phase elution conditions, detection wavelength: 360 nm, flow rate: 1 mL/min, column temperature:30 ℃, sample volume: 10 μL, the isoquercitrin and hyperoside of commercially available fresh hawthorn and the crude slices were detected, and the system suitability was studied. Results Then a study was given to verify the established method, the results showed that the linear range of isoquercitrin and hyperoside were 0.60-38.52 μg/mL and 0.60-38.56 μg/m L;the correlation coefficients were higher than 0.999;and the precision and repeatability of RSD were smaller than 2%, the recovery rate was 101.88%-102.44%, RSD less than 2.5%. Conclusion The method was rapid, simple, accurate, reliable and could be used to detect the isoquercitrin and hyperoside of hawthorn.
