CD4+CD25+调节性T细胞对人外周血内皮祖细胞增殖、迁移、黏附的影响

摘    要:目的:研究CD4+CD25+调节性T细胞对体外培养的人外周血内皮祖细胞(EPCs)增殖、迁移、黏附的影响。方法:密度梯度离心法分离培养人外周血单个核细胞,经FITC-UEA-I和Dil-acLDL双染色鉴定为正在分化的EPCs。进一步采用流式细胞仪检测其表面标志CD34、CD133。磁性细胞分离器(MACS)分离CD4+CD25+调节性T细胞及CD4+CD25-T细胞。将EPCs分别与CD4+CD25+调节性T细胞或CD4+CD25-T细胞共培养36h。采用MTT比色法、transwell小室、细胞计数法观察EPCs增殖、迁移、黏附能力。结果:与对照组和CD4+CD25-T细胞相比,与CD4+CD25+调节性T细胞共培养EPCs增殖、迁移、黏附能力显著增强,且呈浓度依赖性增强。结论:CD4+CD25+调节性T细胞可显著促进EPCs增殖、迁移、黏附能力,此作用可能是其抗AS作用机制之一。
